Abstract:

We describe the proper application of several statistical methods that we have found useful in assessing the quality of biomarkers. Included in the discussion are methods that can be used if the biomarker is dichotomous (e.g., expression of a tumor-associated antigen) or continuous (e.g., body-fluid levels of cotinine). The focus of the discussion is on establishing the reliability and validity of a biomarker, and includes such methods as coefficient kappa, intraclass correlation coefficient, Lin's coefficient of concordance, and the Bland-Altman methodology. We provide guidelines for determining when the various methods should be used, and the methods are illustrated on datasets taken from the biomarker literature.
