Abstract:

Gene expression is controlled by DNA binding proteins whose chromosomal locations are experimentally and computationally difficult to identify. We demonstrate that application of Wilcoxon Rank Sum tests to data generated from hybridizing DNA, which is enriched in regions where these proteins bind, to Affymetrix human genome tiling arrays identifies real binding sites. The array detected regions reveal statistically significant enrichment of consensus sequences and proximity to 5' exons and CpG islands. We discuss the implications this approach has on characterization of gene regulatory networks in human cells.
