Abstract:

Agrobacterium-mediated transformation is the most popular method currently in use for generating transgenic plants. However, very little is known about plant factors involved in this process. Suppressive subtractive hybridization and DNA macroarray technologies were used to identify plant genes that are differentially regulated during the initial stages of Agrobacterium-mediated transformation. An analysis of variance (ANOVA) model and bootstrapping method were employed for the purpose of testing the gene expression changes among different Agrobacterium strainS and uninfected cells.The genes that have different expressions among different cell samples were identified and verified by Northern Blot. A comparison of plant response to different strains of Agrobacterium indicates that transfer of both T-DNA and Vir proteins modulates the expression of host genes during the transformation process.
