Title: Case-sibling Gene-Association Studies for Diseases with Variable Age at Onset

Abstract:

Many complex diseases have variable age at onset. However, standard analytic methods for family-based studies--designed to adjust for possible confounding by population stratification--are based on a binary disease model and consequently can be inefficient or biased. This talk reviews methods for analyzing censored age-at-onset data from family studies, including a novel "retrospective likelihood," which can both test for allelic association and estimate allelic relative hazards. The retrospective likelihood can be more efficient and accurate than standard methods but requires a reasonable external estimate of the baseline hazard.
