Abstract:

This paper describes two methods (jackknife and correlation) of identifying genes that are associated with colon cancer sensitivity to a drug, 5-FU. The sensitivity of each of 30 colon cancer cell lines was quantified. Next, the correlations between 5-FU sensitivity and gene expression were computed for each of 3,725 genes. The N genes with the highest correlations were selected. Then, in another analysis, the jackknife method was applied separately to each gene by leaving out one of the cell lines and using the remaining 29 to build a regression model for sensitivity vs. gene expression. The predicted sensitivity of the "left out" cell line was then computed via the model. This process was repeated for each of the 30 cell lines, and the sum of squared distances between predicted and observed values was computed. N* genes which have the smallest sum square of distances were selected. The two lists of N and N* genes were compared to determine which genes appeared on both lists.
