Abstract:

The problem of simultaneously testing noninferiority and superiority is defined in terms of a multiple test in a continuous family of null hypotheses. Depending on the choice of the noninferiority and superiority margins, sample sizes necessary to achieve a certain power may differ substantially between the hypotheses. One may choose different types of rejection boundaries for the different null hypotheses, e.g., O'Brien and Fleming boundaries for superiority and Pocock boundaries for superiority. This allows a high power with early rejections for the larger margin, whereas for the smaller margin later rejections will be more frequent. Different scenarios for two-stage designs are considered and the relationship to confidence intervals are discussed. An improvement of repeated confidence intervals is given in trails "with a maximal goal." The extension to adaptive sample size reassessment is described.
