Differential expression of proteins between tissues or cell types under different treatment is often assessed by using 2D gel technology. In such experiments proteins are extracted from differently treated tissues and then are subjected to charge and weight separation in a gel-type media. After staining, each protein has its own position on a rectangular gel corresponding to the protein's electric charge and molecular weight. The amount of protein (protein expression) is calculated from the size and the intensity of the spot. In an ideal experiment where each treatment is replicated on a separate gel all spots (proteins) would be clearly identifiable and statistical test would be conducted to determine which spots (proteins) have differential quantities between two or more types of cells. Proteins could be roughly classified in four groups according to their behavior in tissues under different treatments.
