Abstract:

Interaction is used in epidemiology to describe a situation in which two risk factors modify the effects of each other. Case-only design is more powerful to estimate interaction compared with case-control design under the assumption of independence between two genes. The independence assumption is difficult to check based on the available data and violation of which typically leads to inflated type 1 error and loss of power. We will discuss estimation of gene-gene interaction based on case-only data without assuming independence using the Bayesian approach. Some simulation and real data experience with this approach will be presented.
