Abstract:

One question that plagues researchers in the micro-array analyses field is how many replicates are needed to be "sure" that the genes identified as being different are indeed different. Traditional sample size calculations based on Type I error and Power result in very large numbers and are generally useless. In this paper we introduce sample size determination techniques based on the false discovery rate (FDR) and false negative rate (FNR). Sample size calculations are based on maximizing a new form of power (defined as 1-FNR) or exceeding a specified threshold for a measure called correct rejection rate (CRR) that is introduced in this paper. Methods are derived for single-step procedures that do not control FDR but are preferred by most scientists. Extensions to the more complicated step-up and step-down procedures are also discussed.
