Title: MAXIMA: Exact Tests for Linkage Analysis

Abstract:

A new exact test statistic, Max, for sharing two alleles identity by descent (IBD) by affected sib pairs is presented. The test statistic, Max, is the maximum occupancy number based on the equiprobable multinomial distributions with four cells. The reverse-cumulative distribution function, which is used for P-values computation and moments of Max, are exact and derived by combinatorial mathematics. The test, Max, is also extended to multilocus models. Problems concerning comparisons of power between new methods based on Max and the existing tests, including the maximum lod score test by Risch (1990), the mean test, the two-allele test, and the Pearson chi-squared goodness-of-fit test, are considered.
