Abstract:

High-throughput screening has become very popular in the pharmaceutical industry. Robotics and other technical improvements have allowed investigators to screen thousands of potential drug compounds looking for activity in various assays. A problem that plagues these investigators is where to set the cut-off that separates activity from non-activity. When the distribution of the data from a screening activity is examined, invariably the distribution is skewed--many of the compounds show little or no response while a few show a response, often very extreme. We show robust methods, with the correct score function, and implemented using the software RGLM, which can be used to effectively set the cut-off.
