Abstract:

Some genes that affect development and behavior in mamals are known to be imprinted. Hence, incorporating an imprinting parameter in linkage analysis may increase the power to detect linkage for these traits. Here we propose a variance components-based model for testing linkage between a quantitative trait locus and a polymorphic marker in presence of genetic imprinting. We also incorporated sex-specific recombination fractions in this model. We discuss the effects of imprinting and non-imprinting on the power of usual variance components method and on the variance components method with an imprinting parameter. We also provide noncentrality parameters that can be used to determine the required sample size to attain a specified power for a given significance level, which is useful while planning a linkage study. Optimal strategies for a genome scan of potentially imprinted traits are discussed.
