Within the field of clinical cancer research, discovery of biomarkers and genetic mutations that are potentially predictive of treatment benefit are motivating a paradigm shift in how cancer clinical trials are conducted. In this talk, I will provide an overview of basket and umbrella trials, which are increasingly popular design solutions for the study of novel targeted agents. For each, I will describe standardized terminology, discuss scientific and practical advantages and limitations, and provide a detailed real-world example.
