We describe a study to evaluate the impact of Wolbachia suppression targeting Ae. aegypti mosquitoes on reducing the incidence of arbovirus infection in residents of Ponce, Puerto Rico, a historical hotspot for dengue, chikungunya, and Zika outbreaks on the island. We will discuss the choice of a matched cluster-randomized design and summarize existing methods for this design. Finally, we present an approach for estimating the effect of the intervention as an extension to the work of Donner and Klar (2000) and demonstrate how we incorporate covariates into the analysis.
