Abstract:

Physical activity is an important risk factor for a wide range of diseases. Previous genome-wide association studies (GWAS) identified a limited number of genetic loci associated with habitual physical activity and provided evidence that brain tissues may be involved in modulating the genetic effects. These studies, however, are based on self-reported data or on a limited set of physical activity phenotypes derived from accelerometry. We derived 27 physical activity phenotypes from wrist accelerometry data obtained from 93,745 UK Biobank study participants. Single-variant association analysis based on mixed-effects models identified 5 novel loci that were not detected by previous studies of physical activity, sleep duration and self-reported chronotype. Transcriptome-wide association analysis identified one additional novel locus, increasing the total number to 6. Various follow-up analyses indicated the role of the blood/immune system in modulating the genetic effects and a secondary role of the digestive and endocrine systems.
