Title: NanoStringDiffWeb: a Web-Based Tool for Differential Expression Analysis of NanoString NCounter Data

Abstract:

The advanced medium-throughput NanoString nCounter technology provides a novel approach for miRNA profiling and targeted mRNA expression analysis and has been increasingly used for biomarker discovery for early detection, prognosis and prediction. We have previously developed a statistical method, named NanoStringDiff, for differential expression analysis using NanoString data. NanoStringDiff considers a negative binomial model and incorporates data normalization in the model framework. The method has been shown to perform better than other methods such as t-tests. In this poster, we present a new web application named NanoStringDiffWeb. We have improved the NanoStringDiff algorithm by developing an approximation method to calculate the likelihood function of NanoStringDiff, which greatly improves the computational efficiency while maintains the accuracy. We also have introduced several quality control and data visualization plots. Based on the R Shiny platform, the NanoStringDiffWeb provides a user-friendly web interface for biomedical researchers to analyze NanoString data.
