Abstract:

Imaging biomarkers such as FDG-PET standardized uptake value (SUV) require injection of a specific tracer prior to scanning, so analysis of retrospective cohorts is not available to the same degree as for many tissue assays. This limits the scope of technical validation and clinical validation studies of these biomarkers, requiring efficient use of prospective resources and available data. This presentation reviews existing and proposed efforts for the technical validation of the FDG-PET SUV for use as an early response biomarker in oncology studies. Accumulated research data may be used to develop algorithms to harmonize SUV measurements obtained using different scanner models. De-identified clinical scans may be used to identify harmonization priorities and to quantify the range of scanner characteristics to be accommodated.
