We examine tests for binary data subject to order restrictions where the response is umbrella ordered, in which there is a nondecreasing dose response up to some dose level after which there is nonincreasing dose response up to the highest dose level. This situation might be expected, e.g., in a dose-finding study if there is increasing efficacy from the drug up to a certain dose, after which toxic effects overtake the efficacy. This might be a reasonable assumption in some preclinical animal or toxicology studies or dose-finding clinical trials. We propose two new tests for this scenario: a contrast-based test and a quasi-likelihood ratio test. We utilize the E+M method of Lloyd (2008) to make these tests exact the which make the tests more appropriate for small samples. We also examine an exact version of a test by Peddada (2001) for umbrella orders. We compare the power of the three approaches for a variety of scenarios.
