Title: Modified Toxicity Probability Interval (TPI) Design for Phase I Oncology Trials: A Case Study

Abstract:

In this poster, we propose to use a modified version of toxicity probability interval (TPI) method to guide dose escalation in a phase 1 oncology trial. The TPI framework focuses on posterior intervals of the toxicity curve rather than only the point estimate, therefore would have better operating characteristics than some of the point estimate based alternatives. Overdose control is also in place to avoid patients being exposed to high toxic dose levels. Simulations are done to demonstrate the advantage of our proposed method and discussions are made on the appropriate choices of methods in different situations.
