Abstract:

State-space model is proposed to generalize adaptive Bayesian designs for early phase oncology clinical trails, which include, but not limited to, continual reassessment method (CRM), escalation with overdose control (EWOC), and toxicity posterior interval (TPI). The flexibility of state-space model also allows researchers to investigate new adaptive designs within the same framework. An example is given to extend TPI to be fully Bayesian parametric. Estimation of model parameters is made simple by the usage of Monte Carlo simulation, which can be easily implemented and integrated into existing software packages. Its usage is successfully demonstrated by replicating the parameter estimation results from the EWOC's original paper, which resorts to complex numerical integrations.
