Within-tumor heterogeneity has long been a focus in characterizing cancers and identifying treatments. Recent advances in fluorescent in situ hybridization (FISH) has made it possible to examine copy numbers at cellular level and analyze diversity within tumor cells. Statistical issues arise in the estimation of diversity since the total number of types is not observable. In this poster we discuss the biases in Shannon's index and Simpson's index, two popular measures of diversity, due to the total number of types being unobservable. We also provide a maximum-entropy estimator based on the assumption that unobserved types are not more prevalent than the observed types. Data from a recent publication on the diversity of in situ and invasive breast tumors will be used for demonstration
