Abstract:

To study the genetic reasons of tumor growth, cancer formation, and genetic diseases, researchers now use advanced array technologies to conduct DNA copy number experiments as it is believed that cancer development and genetic diseases are usually associated with DNA copy number changes or copy number variations (CNVs) in the genome. It turns out that identifying boundaries of DNA copy number variations along a chromosome or a genome can be viewed as a change point problem of detecting the break-points or boundaries of changes presented in the genomic data. We propose to use a homogenous compound Poisson change point model to characterize the loci of biomarkers and the log ratio intensities and to detect the loci of CNVs in such a change point model. Several applications of the proposed method to the analysis of fibroblast cell line data and the breast cancer/tumor data will be given.
