Tumor sample is now routinely collected in cancer clinical trials. It is hypothesized that the gene expression profile of pretreatment cancer tissue possess the information about patient's disease prognosis and the sensitivity to cancer therapy, and multigene signature predictors of response can be developed, and be used for tailoring individualized treatment. In building supervised survival prediction model, informative genes selection, and methods of synthesizing information across the selected informative genes play a key role. In this presentation, we propose a systematic method in determining informative genes included in model building, and some rule in determining the number of components retention in the survival prediction model.
