Abstract:

The EffTox model is a Bayesian outcome-adaptive method used for determining the best dose of an experimental drug based on desirability, quantified in terms of the trade-off between efficacy and toxicity. In order for this adaptive design to be effective, clinicians must have input on not only the priors, but the acceptability regions that help define desirability. Thus, a firm understanding is of how the model works is needed by the clinicians. This talk will focus on the presentations that have been used to help immunologists and rheumatologists understand the model and then to elicit appropriate information out of them so that the model can be defined. This information includes: the toxicity and efficacy endpoints, their respective expected probabilities of response at different dose levels, and definitions of the acceptability boundaries and tradeoff contours.
