I will describe a Bayesian approach to adaptive randomization in phase II oncology trials, both theory and practice. I will apply these ideas to factorial designs in the early disease setting, including seamless phase I/II trials that address both toxicity and efficacy in the context of combination therapies. Treatment combinations (including single agents) that are not performing well get assigned fewer patients and are eventually dropped. I will describe some of our experiences at M.D. Anderson Cancer Center with these types of designs, including in leukemia and breast cancer.
