Abstract:
|
Immunological correlates of protection are threshold levels of immunological assay values above which individuals are considered protected from disease; more generally, the topic is concerned with the relationship between values of immunological assays and disease, whether characterized as a threshold or a continuous function. Despite considerable effort, the surrogate endpoint/Prentice approach has yielded little in the way of useable results in this area. Correlates of protection have been established for most vaccine-preventable diseases, yet in only two instances have quantitative methods formed a basis for the level chosen. We argue that there are technical reasons why standard approaches are unlikely to yield useful results, and that methods more specifically geared to the problem are needed. Progress towards such methods will be presented and current challenges outlined.
