Abstract:

Results from two recent collaborative efforts to examine the value of single nucleotide polymorphisms (SNPs) for postmenopausal breast cancer risk prediction will be briefly described. One of these involved about 2000 breast cancer cases and controls, and formed a risk estimate by multiplying the Gail model absolute risk estimate by a 7-SNP odds ratio factor.The second was a larger cohort collaboration involving about 6000 cases and controls, developed a joint model for clinical vaiables and 10 SNPs.Both concluded that SNPs offer some modest improvement in risk classification.Comments will be provided on prediction model evaluation and risk attribution.
