Abstract:

Adaptive Phase II/III clinical trials incorporating treatment selection have gained increasing interest in the past few years. In these designs, several treatments are compared to a control in a first stage. After an unblinded interim anaysis one or more treatments are selected for the second stage and sample sizes may be reassessed. The final analysis is based on the data of both stages. For such designs, we propose adaptive, sequentially rejective test procedures defined by directed, weighted graphs that control the family wise error rate in the strong sense. Control of the Type I error rate is guraranteed even if the adaptation rules are not pre-specified. These procedures generalize the recently proposed fixed sample, graph based multiple testing procedures to adaptive tests and rely on adaptive weighted Bonferroni Holm tests.
