Abstract:

Population-based case-control design and family-based case-parents design are frequently used to investigate association between a candidate locus and disease. Both designs have advantages and limitations. However, one often collects genotype data from family triads, and unrelated subjects in association studies. Under such situations, it is desirable to have an association test which is not only robust to the effect of population stratification but also more powerful than the usual tests based on case-parents data alone. In this paper, we propose a two-stage association test based on combining genotype data from family triads, unrelated cases and/or controls. This test is robust to the genetic model and effect of population stratification, and allows for missing parental data.
