Abstract:

ABT-263, a potent, orally bioavailable Bcl-2 family protein inhibitor that we have shown to be effective against small cell lung cancer (SCLC) and leukemia/lymphoma cell lines, is in multiple Phase 1/2a clinical trails for B-cell lymphoma, CLL and SCLC. This talk will cover the identification of key Bcl-2 family proteins and those outside the Bcl-2 family that influence the cellular response of SCLC and leukemia/lymphoma cell lines to ABT-263. Special emphasis will be on the analysis strategy used for deriving gene signatures, starting from the filtering of genes from the genomics data set to gene signature derivation to the use of classification algorithms and finally, the evaluation of predictive performance of this entire process using internal cross-validation. Plans for incorporating the use of these genes in clinical trials and as patient stratification markers will be outlined.
