Abstract:

In case-control study identifying disease susceptibility loci, it has shown that the interaction caused by single nucleotide polymorphisms (SNPs) at unlinked genes plays an important role to influence risk of a disease. A novel statistical approach is proposed to detect gene-gene interactions at the allelic level contributing to a disease trait. With a new allelic score inferred from the observed genotypes at two or more unlinked SNPs, I test for association of the allelic scores with a disease trait based on a score test derived from logistic regression and a F test from Armitage regression. Based on non-centrality parameter approximation of a score test, the analytical powers and type I error rates over 6 two-interaction models are investigated. Simulation studies demonstrate that the power of the score test is asymptotically equivalent to that of the F test by Armitage method.
