Abstract:

We discuss statistical analyses for time course studies using microarrays which arose in the CardioGene study. CardioGene is a prospective epidemiological study of restenosis in bare metal stents for the treatment of coronary artery disease. 350 patients were prospectively enrolled at the time of stent implantation, and the primary clinical outcome was determined 12 months after stent placement. Blood was sampled prior to stent placement and afterwards at 2 weeks and 6 months, and mRNA gene expression profiles obtained using Affymetrix U133A arrays. We will describe statistical methods we used to identify differential expression patterns over time between patients who developed restenosis and those who didn't. Some discussion on further research topics such as validation of the findings and integrated analysis of the gene expression profiles and the DNA genotype data will be provided.
